Can a brake vacuum leak cause the 3 amigos in a Disco2

The brake booster vacuum leak you are causing by moving the pipe should not cause the 3 Amigos. The brake system does not monitor brake vacuum level - so will not "know" the leak is happening.
The leak you are creating is only temporary while you're distorting the seal around the vacuum non return valve. All vehicles will leak vacuum if you move the vac. pipe - so it's not something to worry about.
The warning lights coming on when you operate the brake is a separate problem.
Ideally you need to get access to a fault code reader to understand what fault or faults are causing them - as there are many potential causes & changing each item until you find the cause could waste a lot of money.
Add your location & you may find someone local who will volunteer to connect their diagnostic kit to view the fault codes.
