When engaging reverse and moving backwards there is a quite loud whining noise. Don't think this is normal. Could it be a transmission issue?
No noise in the forward gears

shouldnt be any whine in reverse gear as r380 uses helical cut gears for reverse unlike lt77,check the condition of oil in both boxes by draining some out

Spigot bush? I have same problem, I was advised this is common problem, cheap part, but gearbox off to do the job, I'm leaving mine till clutch needs replacing, as been told won't do any harm, just annoying to hear,

The spigot bush would affect all gears, and as the first motion shaft is so long, even if it was missing, the impact at the gearbox end would be small. It would make the clutch engagement very jerky though as the plate wouldn't be centralised.

Reverse gear whine does sound like something in the bearings is not too clever, and that would also go for the transfer box gears too, which are helical and biased/thrust one way when going forwards and the other way when going backwards.

My suggestion is the same as James above, drop some oil out of both boxes and see what comes out in the way of debris, I'd lean slightly towards a tranny box problem.

spigot bush is a common issue with td5 and can be shown as it makes a noise when pedal is at slipping point but doesnt when clutch is fully depressed hence often noticeable when reversing as clutch is not usually fully engaged, but will make the same noise in forward gears too

Exactly what mine does do,
