The Defender Parts Catalogue doesn't list a screw to hold the lock barrel in place, it's a small pin that you have to depress to remove the barrel. If yours had a screw fitted I can only presume that it was fitted by a previous owner.

If the rest of the exhaust is ok just replace the rotten section. Have a look at Paddocks or similar parts supplier for it. It's easy to replace the section, just a couple of bolts ( which will have rusted solid so cut them off ), buy new bolts, the parts supplier will have them...

If it's a coil sprung chassis it's not a 1976 88 inch Land Rover so your V5 is invalid.
