newbie wanting advise AGAIN!!

OK so the Merc into the disco is a no go im not experienced enough
SO NOW , back down to earth , when i pull away i get a kinda take-up clonk noise, much more apparent when going to reverse. so my question is to all you experienced disco owners
IS IT DIFF
IS IT GEARBOX
IS IT TRANSFER BOX
im running a 2.5tdi and i do have a complete 3.9 v8 for spares
any help will be gratefully accepted
